B7-H3 (also known as CD276) is a member of the B7 superfamily of immunoregulatory proteins. It is a type I transmembrane glycoprotein that is expressed in various human tissues and upregulated in numerous cancers, including lung, breast, prostate, and colorectal tumors. B7-H3 plays a complex role in immune modulation and has been shown to both stimulate and inhibit T cell responses depending on the context. Recent studies suggest that B7-H3 contributes to tumor immune evasion, angiogenesis, and metastasis, making it a promising target for cancer immunotherapy. Due to its restricted expression in normal tissues and high expression in tumors, B7-H3 is being actively investigated in antibody-drug conjugates (ADCs), CAR-T therapies.
